Effect of Cultural Conditions on Rhizome Growth and Organogenesis of Cymbidium lancifolium Native to Korea In Vitro


Abstract
The experiments were attempted to investigate the effect of various hydrogen ion concentration of medium, length and density of rhizome, activated charcoal, light or dark and temperature condition in vitro on rhizome growth and shoot formation of Cymbidium lancifolium native to Korea in vitro.
The rhizome growth was more promoted on pH 5.5 and 1.5§¯ length of rhizome segement innoculated 100§¢ Erlenmeyer flask under light condition at 19¡É
However the shoot and root formation were most effective under light condition at 25¡É in the medium adjusted pH 4.5 and one or two rhizome segements cultured per 100 §¢ Erlenmeyer flask. Rhizome growth, shoot and root formation were promoted with addition of 1.0 g/§¤ activated charcoal in modified Murashige and Skoog medium.
